Russian scientists have discovered that drugs used to treat alcoholism and cancer are also effective against SARS-CoV-2.
According to the magazine Mendeleev Communications, it became clear to researchers from the School of Higher Economics and the Institute of Organic Chemistry of the Russian Academy of Sciences, that some drugs used in the treatment of alcoholism and cancer are also effective against the emerging coronavirus.
The magazine notes that it usually takes 4-7 years to demonstrate that the drug is not dangerous experimentally. But the pandemic of “Covid-19” continues to spread, so doctors and medical experts are searching for a drug that is used successfully in treating other diseases, so it may help in the treatment of “Covid-19” as well.
Russian chemists have performed extensive calculations using a supercomputer to find an effective drug. They chose the Mpro protein that is necessary for the reproductive pathogen to reproduce. This same protein is found in different strains of the SARS-CoV-2 virus. That is, changes in the gene encoding this protein are potentially lethal to the virus. Therefore, any new strains that appear in the future remain the same Mpro protein, and the anti-drug works effectively.
Scientists usually search for a substance that is able to bind to the active center of the protein by molecules that perform the functions of this protein and block its activity. Since the active center of Mpro is known, but there is no certainty that there are no other centers, because proteins can contain tens and hundreds of active centers.
Based on this, the researchers chose the required material according to their new method which they called the “surface fusion method” that the computer set on the surface of the protein molecules, not just its active center.
After testing a group of drugs, the researchers chose two drugs: the first – disulfiram, which is used to treat alcoholism, and the second – neratinib, which was recently approved to treat breast cancer. The results of previous tests showed the effectiveness of the drug disulfiram in the treatment of SARS-CoV and MERS, close to the current pandemic.
Chemical experiments show that this drug successfully blocks the Mpro protein, while the Neratinib drug, its effectiveness in this field is not at the required level. The clinical trials of the two drugs will begin on the first of next September.
And if the results of clinical tests confirm the effectiveness of the drug disulfiram, it will become an effective tool in the fight against coronavirus, and at the same time may reduce the desire to drink alcoholic beverages.
It should be noted that Chinese scientists have reached the same results regarding the effectiveness of the drug disulfiram.
